Mylonib contains Ruxolitinib, a JAK1 and JAK2 inhibitor that targets overactive signaling inside the bone marrow. JAK (Janus kinase) enzymes relay the growth signals that control blood cell production and immune activity. Doctors prescribe Mylonib for adults with myelofibrosis, a disorder where scar tissue replaces healthy marrow.

Mylonib also treats polycythemia vera in adults who cannot tolerate or no longer respond to hydroxyurea. Specialists set the dose by platelet count, often starting at 15mg twice daily. Patients swallow one tablet twice daily, with or without food, exactly as the haematologist directs.

How Does Mylonib Work?

Mylonib works by blocking JAK1 and JAK2, two enzymes in the JAK-STAT signaling pathway inside blood cells. In myelofibrosis and polycythemia vera, mutations such as JAK2 V617F keep this pathway switched on permanently. Ruxolitinib quiets the pathway, lowering inflammatory cytokines and slowing the overproduction of blood cells.

Mylonib reduces an enlarged spleen and eases symptoms such as night sweats, fatigue, and weight loss. By dampening JAK signaling, Ruxolitinib lowers phosphorylation of STAT proteins that drive abnormal cell growth. In phase 3 trials, Ruxolitinib shrank spleen volume by 35% or more in many patients at 24 weeks.

Mylonib Side Effects

Mylonib commonly lowers blood counts and can raise infection risk, so patients need regular blood monitoring. Report fever, unusual bruising, or any sign of infection to your haematology team without delay.
